Lipid metabolism of Ehrlich ascites tumor cells grown in chemically defined albumin medium.

چکیده
Total lipids of Ehrlich ascites tumor cells grown in nutrient medium supplemented with 1% bovine serum albumin instead of 15% native horse serum and rate of incorporation of [3H] acetate into total lipids are reduced, while the cholesterol content of the cells is higher than of controls grown in normal medium. The lower lipid content of cells cultured in albumin supplemented medium is explained by a lack of exogenous lipids, which are normally taken up from the serum containing nutrient medium. A several fold increase of incorporation of labelled choline and ethanolamine into the phospholipids of the cells grown in the modified medium is observed. We suggest, that the observed stimulation of the incorporation of phospholipid precursors reflects changes in the dynamic state of the membranes produced by serum depletion.

Proliferation and metabolic activities of Ehrlich ascites tumor cells in chemically defined albumin media.
Cell proliferation, viability, DNA-, RNA-, protein synthesis, amino acid transport, repiration and lactate/glucose quotient of Ehrlich ascites tumor cells grown in suspension culture in serum free medium supplemented with albumin charges of different origin were studied. Optimal cell growth was obtained in nutrient medium supplemented with 1% bovine serum albumin (Cohn-fraction V, Serva). Cell ...

Cell proliferation, cell cycle distribution, viability, uptake of alpha-aminoisobutytic acid, cyclo leucine and 2-deoxyglucose as well as DNA, RNA and protein synthesis of Ehrlich ascites tumor cells grown in suspension culture over three passages in serum free nutrient medium supplemented with 1-2% bovine serum albumin) Cohn fraction V, Serva) were measured and compared to cells grown in mediu...
متن کاملRelease of free fatty acids from Ehrlich ascites tumor cells.
Ehrlich ascites tumor cells release free fatty acids (FFA) during in vitro incubation in media that contain albumin. The released FFA are derived by lipolysis from endogenous lipid esters. Addition of glucose to the incubation medium greatly decreases the quantity of fatty acid released by the cells. Cyanide, which inhibits endogenous lipid oxidation but not lipolysis, increases the quantity of...
متن کاملEffect of liposome-encapsulated quercetin on DNA synthesis, lactate production, and cyclic adenosine 3':5'-monophosphate level in Ehrlich ascites tumor cells.
Quercetin (3,3',4',5,7-pentahydroxyflavone) entrapped within phospholipid vesicles inhibited [3H]thymidine incorporation and lactate production in Ehrlich ascites tumor cells. The presence of serum proteins in concentrations of up to 40 mg/ml did not alter the extent of the inhibitory effect of the liposome-entrapped flavone. At the same concentrations, serum proteins eliminated almost complete...
متن کاملInfluence of pH of the medium on free fatty acid utilization by isolated mammalian cells.
Studies with Ehrlich ascites tumor cells showed that small decreases in the pH of the incubation medium from 7.4 increase the magnitude of incorporation of free fatty acid (FFA) into the cells from an albumin solution. A similar effect occurred when rabbit erythrocytes, rat heart slices, or rat liver slices were incubated with FFA-bovine albumin solutions and when tumor cells were incubated wit...
